| dc.description.abstract | Резюме: В обзора се разглежда ролята на IL-18 във възпалението. Възпалението като защитен механизъм на организма води до освобождаване на различни медиа- тори, включително цитокини. IL-18 е член на IL-1 семейството и най-вероятно играе съществена роля във възпалителния процес. IL-18 действа главно вър- ху Th1 лимфоцитите, продуциращи IFN-γ в присъствието на IL-2 или IL-12, за да се засили тяхната трансформация и преживяемост. В допълнение, IL-18 индуцира продукцията на цитокини и усилва цитотоксичното действие на лимфоцитите. Характерно за IL-18 и IL-1β е, че те се активират от вътрекле- тъчни мултимолекулни комплекси, т.нар. инфламазоми. Инфламазомите кон- тролират активността на каспаза-1, която разцепва неактивните прекурсори про-IL-1β и про-IL-18, за да се произведат биологично активните им форми. В литературата има много малко данни за точния механизъм, обуславящ въздействието на IL-18 върху таргетните клетки, както и ролята на този интерлевкин във възпалението. Във връзка с това в настоящия обзор се фокусираме върху механизмите, обуславящи ефекта на IL-18 върху лимфоцитите, както и върху биологичната и патологичната роля на IL-18 в сравнение с другите членове на IL-1 семейството. ***** Abstract: This review focuses on the role of IL-18 in inflammation. The inflammation as a protective response of the body leads to the release of inflammatory mediators including cytokines. IL-18 is a member of the IL-1 family and most likely plays essential role during the inflammatory reactions. IL-18 acts on Th1 lymphocytes that are mainly producing IFN-γ in the presence of IL-2 or IL-12 to enhance their augmentation and also their survival. In addition, IL-18 induces production of cytokines and increases cytotoxic activity of lymphocytes. Typical of IL-18 and IL-1β is that they are activated by intracellular multi-molecular complexes, called inflammasomes. Inflammasomes control activity of caspase-1, which cleaves the inactive precursor pro-IL-1β and pro-IL-18, to produce the biologically active forms. In the literature there is a little bit of information on the exact mechanism underlying the efficacy of IL-18 on its target cells and the role of this interleukin in inflammation. Because of this, in this review we focus on the mechanisms underlying the effect of IL-18 on lymphocytes as well as on the biological and pathological role of IL-18 in comparison with other members of the IL-1 family. | en_US |
